摘 要:目的研究己酮可可碱对兔实验性蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后脑脊液中TGF-β1的影响及其与蛛网膜纤维化的关系。方法新西兰兔55只,随机分为空白组、模型组、己酮可可碱干预组,应用枕大池两次注血法建立兔SAH模型,在模型组基础上给予己酮可可碱干预,分别在第3、7、11、16、21天检测各组脑脊液中TGF-β1的浓度,第21天处死动物取脑,Masson法染色后,应用图像处理软件显示胶原纤维含量。结果模型组和干预组脑脊液中TGF-β1的浓度在第3天和16天出现两个高峰,但干预组较模型组显著降低(P<0.05),空白组无明显改变。Masson染色显示模型组蛛网膜下腔纤维含量明显增多,与干预组和空白组相比差异具有显著性,干预组蛛网膜下腔纤维含量较空白组增多,较模型组减少,差异均具有显著性(P<0.05)。结论己酮可可碱能降低兔实验性蛛网膜下腔出血后脑脊液中TGF-β1浓度,具有一定的抗蛛网膜纤维化作用。Objective To study effects of pentoxifylline on rabbit′s cerebrospinal fluid TGF-β1 with experimental subarachnoid hemorrhage and its relationship with arachnoid fibrosis. Methods 55 New Zealand rabbits were randomized into blank group,model group,PTX group.The blood infusion twice into cisterna magna was applied to establish the rabbit SAH model,in the model group given pentoxifylline intervention,on 3,7,11,16,21 days detected in the cerebrospinal fluid of TGF-β1 concentration,twenty-first day,animals were sacrificed and brain,Masson method of dyeing,the application of image processing software to display in collagen fiber content. Results Model group and PTX group in cerebrospinal fluid of TGF-β1 concentrations in the third and 16 day appeared two peaks,The intervention group was significantly lower than those in the model group(P0.05).Blank group showed no change.Masson staining showed model of subarachnoid cavity fiber content increased significantly,compared with the intervention group and blank group,the difference was significant.The subarachnoid fiber content of intervention group was increased compared to blank group,but reduction compared to model,differences were significant(P0.05). Conclusion Pentoxifylline can reduce TGF-β1 concentration in cerebrospinal fluid of rabbits after experimental subarachnoid hemorrhage,and has certain effect of resistance on arachnoid fibrosis.
