绞股蓝皂甙诱导人口腔鳞癌颈淋巴结转移癌细胞凋亡的研究

摘　　要：目的　探讨绞股蓝皂甙诱导人口腔鳞癌颈淋巴结转移癌GNM细胞凋亡的发生机制。方法　采用活细胞观察、超微结构观察、流式细胞仪及DNA琼脂糖凝胶电泳进行观察和检测。结果　 5 0 μg/ml的绞股蓝皂甙作用于GNM细胞后 ,细胞出现典型的核染色质凝集、核固缩、破裂 ;用FCM检测到G1峰前出现亚二倍体核型峰 ,DNA琼脂糖凝胶电泳带呈现DNA断裂损伤。结论　绞股蓝皂甙可诱导人口腔鳞癌颈淋巴结转移癌细胞发生凋亡。诱导细胞凋亡可能是其抑癌机理之一。Objective To investigate the mechanism by which gypenosides induce apoptosis in neck lymph node metastatic cell line derived from human oral squamous cell carcinoma.Methods Cell morphology,flow cytometry and DNA agarose gel electrophoresis were performed for gypenosides-induced apoptosis in GNM cell line.Results Typical chromatin condensation,kanyopyknosis and fragmentation were observed when 50 μg/ml 1 gypenosides were added in GNM cell line,sub-G peak was recorded in DNA histogram of FCM,DNA ladder was not viewed on agarose gel.Conclusions Gypenosides may induce apoptosis in GNM cell line,which may be one of mechanisms of its suppressive effect for cancer.
