摘  要:目的:初探炎性刺激对人多核白细胞(PMN)和脐静脉内皮细胞(HUVEC)的血小板内皮细胞粘附分子-1(PECAM-1)表达的影响和PECAM-1的可能作用。方法:使用流式细胞仪和免疫组化法,观察脂多糖(LPS)、白介素(IL)-1β和肿瘤坏死因子(TNF)-a对PMN和HUVEC PECAM-1表达的变化。结果:LPS、1L-1β和TNF-a可引起PMN表达PECAM-1蛋白减少,而对HUVEC PECAM-1的表达无显著影响,但组化显著在HU-VEC连接部的PECAM-1染色变淡。结论:上述炎性刺激物不仅可引起PMN表达PECAM-1减少、激活PMN,而且可改变内皮细胞PECAM-1的分布,因此PECAM-1在炎症中可能起重要作用。Objective: Preliminarily effect of inflammation stimuli on platelet endothelial cell adhesion molecule-1 (PECAM-l) in polymorphonuclear(PMN) and human umbilical vein endothelial cells(HUVEC) and potential roles of PECAM-1 were explored. Method: Changes of PECAM-1 expression in PMN and HUVEC stimulated by lipopolyaccharide (LPS). interleukin-1β (IL-1β) and tumor necrosis factor-α(TNF-α) by flow cytometry and immunohistochemistry were observed. Results: LPS. IL-1β and TNF-α induced PECAM-l protein expression reduction in PMN, but did not affect significantly PECAM-1 expression in HUVEC. Immunohistochemistry re vealed reduced PECAM-1 staining in HUVEC junctions stimulated by inflammation stimuli. Conclusion: Inflammation stimuli above can cause not only reduction of PECAM-1 expression for activating PMN but also PECAM-l redistribution on HUVEC, these imply PECAM-1 maybe play an important role during inflammation.
