高迁移率族蛋白B1在肝癌细胞中的表达及其临床意义

摘　　要：目的比较不同转移能力人肝癌细胞和人正常肝细胞中高迁移率族蛋白B1(high mobility group protein box1,HMGB1)mRNA和蛋白表达的差异,以探索HMGB1在原发性肝癌生长和转移中的临床意义。方法应用PCR法检测高转移人肝癌细胞LM3、低转移人肝癌细胞HEPG2和人正常肝细胞LO2 HMGB1mRNA的表达,Western blot法检测高转移人肝癌细胞LM3、低转移人肝癌细胞HEPG2和人正常肝细胞LO2 HMGB1蛋白的表达。结果高转移人肝癌细胞LM3、低转移人肝癌细胞HEPG2的HMGB1mRNA和蛋白均较正常肝细胞LO2的表达明显升高(P<0.05);而且,高转移人肝癌细胞LM3较低转移人肝癌细胞HEPG2 HMGB1mRNA和蛋白的表达也明显升高(P<0.05)。结论 HMGB1在高转移人肝癌细胞LM3中较在低转移人肝癌细胞HEPG2和正常肝细胞LO2中高表达,提示HMGB1可能与原发性肝癌的生长和转移有关。Objective To investigate expression of high mobility group protein box 1（HMGB1） in human hepatocellular carcinoma cells.Methods PCR and Western blot were used to study HMGB1 gene expression at the mRNA and protein levels in hepatocellular carcinoma cells（LM3 and HEPG2）.A normal cell line（LO2） was used as a control.Results Expression levels of the HMGB1 gene（mRNA and protein） were much higher in hepatocellular carcinoma cells LM3 and HEPG2 than in LO2（P0.05）.Moreover,HMGB1 expression was also higher in the highly metastatic hepatocellular carcinoma LM3 cells than in the less metastatic hepatocellular carcinoma HEPG2 cells（P0.05）.Conclusion High expression of HMGB1 in hepatocellular carcinoma cells suggests that it may play an important role in the growth and metastasis of hepatocellular carcinoma.
