摘  要:目的研究三氧化二砷(As_2O_3)对人、鼠肝癌细胞株凋亡的诱导作用.方法用不同质量浓度(0.25,0.5,1,2,3,4mg/L)的三氧化二砷处理体外培养的鼠7919,人 HePG_2肝癌细胞株,以 MTT 比色法、AO/EB 荧光染色法、DNA 凝胶电泳和免疫组化染色法检测细胞凋亡.结果不同浓度的三氧化二砷均可抑制7919,HePG_2细胞生长;AO/EB 荧光染色在荧光显微镜下观察肝癌细胞呈典型的凋亡形态学改变;DNA 凝胶电泳呈梯形条带为凋亡的特征;免疫组化染色加药组 bcl-2表达下调,bax 表达上调,对照组 bcl-2表达上调,bax 表达下调,两株细胞结果相同.结论 As_2O_3对两株细胞均有诱导凋亡作用.这为临床应用As_2O_3治疗肝癌提供了一个可供参考的实验依据.AIM To investigate the function of arsenic trioxide (As_2O_3) to induce apoptosis of human and mice hepatocellular carcinoma cell lines. METHODS Mice 7919 and human HePG_2 hepatocellular carcinoma cell lines were treated with As_2O_3 at different mass concentrations (0.25,0.5,1,2,3,4 mg/L) in vitro, then apoptosis was observed by MTT assay,AO/EB fluorescence microscopy,DNA agarose gel electrophoresis and immunohistochemical staining. RESULTS As_2O_3 at different concentrations could inhibit the growth of 7919 and HePG_2 cell lines.HCC cells displayed the typically morphological features of apoptosis by AO/EB fluorescence microscopy.DNA ladder of agarose gel electrophoresis was the typically morphological feature of apoptosis.The immunohistochemical staining showed the expression of bcl-2 decreased and bax increased in the experimental group,and the expression of bcl-2 increased and bax decreased in the control group.The results were the same in two cell lines. CONCLUSION As_2O_3 has the function of inducing apoptosis to two cell lines.It is a scientific proof for clinical treatment of hepatocarcinoma by As_2O_3.
