摘  要:目的:研究共刺激分子B7-H1和抑癌基因PTEN在人结肠癌组织中的表达及意义,并探讨B7-H1和PTEN的相互关系。方法:运用免疫组织化学SP法检测46例结肠癌组织、20例腺瘤性息肉组织、28例慢性炎症组织中B7-H1和PTEN的表达情况,并对其表达水平与结肠癌临床病理参数之间的关系进行相关性分析。结果:B7-H1分子在结肠癌组织、腺瘤性息肉及慢性炎症组织中的表达率分别为82.6%(38/46)、20.0%(4/20)、21.4%(6/28)。其阳性表达与患者年龄、性别、癌症分化程度、淋巴结转移及浸润深度未见相关(P>0.05)。PTEN分子在结肠癌组织中、腺瘤性息肉及慢性炎症组织中的表达率分别为54.3%(25/46)、75.0%(15/20)、78.6%(22/28),其阳性表达与患者年龄、性别、淋巴结转移及浸润深度无关(P>0.05),而与癌症的分化程度相关(P<0.05)。B7-H1和PTEN在结肠癌组织中的表达未见负相关性,但临床数据提示二者的表达率呈相反趋势。结论:B7-H1高表达、PTEN低表达在结肠癌发生发展中起重要的作用,他们可作为抗肿瘤治疗的有效新靶点。Objective:To study the expressions of co-stimulatory molecule B7-H1 and tumor suppressor gene PTEN in human colon carcinoma tissues and the significance,and explore the relationship between B7-H1 and PTEN.Methods: The immunohistochemical SP method was used to detect the expressions of B7-H1 and PTEN in 46 cases of colon carcinoma tissues,20 cases of adenomatous polyp tissues and 28 cases of chronic enteritis tissues,then analyzed the relationship between the expression level and clinical pathology parameters.Results: The expression rates of B7-H1 molecules in colon carcinoma tissue,adenomatous polyp chronic enteritis tissues were 82.6%(38/46),20.0%(4/20) and 21.4%(6/28) respectively,and the positive rate had no relationship with patients' age,gender,cancer differentiation,lymph node metastasis and invasion depth(P0.05).The expression rates of PTEN molecules in colon carcinoma tissue,adenomatous polyp chronic enteritis tissues were 54.3%(25/46),75.0%(15/20) and 78.6%(22/28) respectively,and the positive rate had no relationshiop with patients' age,gender,lymph node metastasis and invasion depth(P0.05),but the cancer differentiation(P0.05).The correlation between B7-H1 and PTEN in colon carcinoma tissue was not negative(P0.05),however,the counter trend of their expression rate was showed from the clinical data.Conclusion: High expression of B7-H1 and low expression of PTEN in colon carcinoma tissue play an important role in the oncogenesis and progression of colon carcinoma,they can be used as an effective new target on antineoplastic therapy.
