摘  要:目的探讨大黄素对大鼠急性水肿型胰腺炎的保护作用机制。方法应用蛙皮素制作急性水肿型胰腺炎模型,将30只大鼠随机分为空白对照组、水肿型胰腺炎组和大黄素治疗组。于造模后12 h,采用酶联免疫法(ELISA)和逆转录-多聚酶链反应(RT-PCR)技术检测血清淀粉酶测(AMY)、白介素-1β(I L-1β)和胰腺组织肿瘤坏死因子-α(TNF-α)水平。结果大黄素治疗组胰腺炎组织损伤明显减轻,治疗组血中IL-1β和胰腺组织中TNF-α的含量显著低于胰腺炎组。结论大黄素可能通过抑制IL-1β、TNF-α的产生,从而减轻水肿型胰腺炎症。Objective To explore the mechanisms of the protective effects of emodin on acute edematous pancreatitis in rats.Methods Acute edematous pancreatitis models of rats were established with bombesin.Thirty rats were randomly divided into control group,edematous pancreatitis group and emodin treatment group.After 12 h of the models developed,the expressions of amylase(AMY),interleukin 1β(IL-1 β) and tumor necrosis factor α(TNF-α) in pancreatic tissues were separately determined with enzyme-linked immunosorbent assay(ELISA) and reverse transcription-polymerase chain reaction(RT-PCR) methods.Results Emodin treatment group significantly reduced pancreatitis tissue injury.The levels of IL-1β in serum and TNF-α in pancreatic tissue in treatment group were significantly lower than those in the pancreatitis group.Conclusion Emodin can relieve acute edematous pancreatitis in rats by inhibition of the production of IL-1 β and TNF-α.
