摘  要:应用Plackett-Burman设计法对影响液体发酵血红铆钉菇菌丝体产量的培养基组分进行筛选,确定影响菌丝体产量的主要因素为蔗糖、蛋白胨和硫酸锌。在此基础上采用最陡爬坡实验结合Box-Behnken响应面法优化血红铆钉菇液体发酵培养基。确定最优培养基配方为:蔗糖45g/L,蛋白胨5.62g/L,ZnSO433.34mg/L,K2HPO41g/L、KH2PO40.5g/L、MgSO40.5g/L。此条件下菌丝体预测产量为9.68g/L,实际试验产量为9.58g/L,证明模型预测具有很好的准确性。The Plackett-Burman design was used to evaluate the effects of culture medium component which could affect the yields of Gomphidius rutilus mycelium,and sucrose,peptone and ZnSO 4 were chosen as the most important factors.Subsequently,path of Steepest Ascent and Box-Behnken response surface methodology were employed to determine the maximum yields of Gomphidius rutilus mycelium.The optimized mediums was as follow:sucrose is 45 g/L,peptone is 5.62 g/L,ZnSO 4 is 33.34 mg/L,K 2 HPO 4 is 1 g/L,KH 2 PO 4 is 0.5 g/L and MnSO 4 is 0.5 g/L.The predicted yield of Gomphidius rutilus mycelium is 9.68 g/L under the optimized conditions,and the practical mycelia yield can reach 9.58 g/L.The model was highly significant and indicated highly precision.
