摘 要:目的:研究并优化大枣环磷酸腺苷(cAMP)的提取纯化工艺。方法:采用所建立的RP-HPLC法测定cAMP含量,通过单因子和正交实验优化提取条件,根据静态吸附和解吸实验筛选大孔吸附树脂纯化cAMP。结果:水提法提取大枣cAMP的最佳条件为料液比1:3、90℃提取4h,提取率为12.03mg/kg鲜枣;从5种树脂中筛选出AB-8大孔吸附树脂纯化cAMP,15μg/mL的cAMP大枣提取液以1BV/h流速上样64mL,吸附24h,30%乙醇以0.5BV/h流速洗脱4h,获得纯度为58.2%的cAMP样品。Objective:To study and optimize the conditions of extraction and purification of cyclic adenosine monophosphate(cAMP) from Chinese jujube.Methods:The RP-HPLC method was used to mensurate the content of cAMP,and the conditions of extraction were optimized with single factor and orthogonal experiment,and the macroporous adsorptive resins were selected to purify cAMP based on adsorption and desorption tests.Results:The optimum extraction conditions for cAMP from jujube by water were solid-liquid ratio 1:3,90 ℃ for 4 h,and the extraction rate of cAMP reached 12.03 mg/kg fresh jujube.In addition the AB-8 was selected from five type of macroporous adsorption resins to purify cAMP,and the purity of cAMP attained 58.2% under the conditions of 15 μg/mL cAMP extract being added 64 mL with flow rate 1 BV/h,AB-8 adsorbing 24 h,30% ethanol eluanting 4 h at flow rate 0.5 BV/h.
