摘  要:目的:探讨脑胶质瘤组织中细胞周期蛋白(Cyclin)D1、Cyclin A、Cyclin B1和核分裂特异性抗体(PHH3)的表达特征及规律。方法:采用免疫组化S-P法检测Cyclin D1、Cyclin A、Cyclin B1和PHH3在59例脑胶质瘤组织(Ⅰ级10例、Ⅱ级15例、Ⅲ级15例、Ⅳ级19例)和37例正常脑组织中的表达情况。结果:Cyclin D1、Cyclin A、Cyclin B1、PHH3在正常脑组织中不表达。胶质瘤组织中Cyclin D1、Cyclin A、Cyclin B1、PHH3的阳性表达率分别为69.5%、69.5%、62.7%和84.7%。4级脑胶质瘤中Cyclin D1、Cyclin A、Cyclin B1、PHH3的阳性标记指数比较差异有统计学意义(P<0.01)。随着胶质瘤级别升高,Cyclin D1、Cyclin A、Cyclin B1、PHH3的标记指数呈升高趋势。结论:通过免疫组织化学法分析细胞周期的数据有利于肿瘤分级,并与预后相关。Objective: To explore the expression and significance of Cyclin D 1, Cyclin A, Cyclin B 1 and anti-phosphohistone-H3 (PHH3) in human glioma. Methods: Expression levels of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 were analyzed by S-P immunohistoehemical analysis in 59 patients with glioma(grade I 10 cases, grade Ⅱ15 cases, grade Ⅲ 15 and grade Ⅳ 19)and 37 normal brain tissues. Results: There were no expressions of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in normal brain tissues. The positive expression rates of Cyclin D1, Cyelin A, Cyclin B1 and PHH3 in gliomas were 69.5%, 69.5%, 62.7% and 84.7% respectively. With the increasing of pathological grades of glioma, the mean labeling indexes for Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in human gliomas increased accordingly. The differences of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 labeling index were significant between four groups (P 〈 0.01). Conclusion: The cell cycle phase can be analyzed by immunohistochemical stain, which is beneficial for tumor grading and helpful to predict the prognosis.
