摘 要:目的:研究黄芪、水蛭、大黄及其复方抑制大鼠系膜细胞(mesangial cells,MC)产生纤维连接蛋白(fibronec-tin,FN)及Ⅳ型胶原(typeⅣcollagen,Col-Ⅳ)的强弱。方法:在大鼠MC体外培养的基础上,将脂多苷糖(lipopolysac-charide,LPS)刺激的MC增生模型分为9组:控制组、模型组、对照组、黄芪组、水蛭组、大黄组、雷公藤多苷组、复方组、复方+雷公藤多苷组。采用ELISA法检测黄芪、水蛭、大黄及其复方等含药血清对MC表达FN及Col-Ⅳ的影响。结果:各含药血清组均能明显降低经LPS刺激后的大鼠MC产生FN及Col-Ⅳ的量,以复方+雷公藤组作用最佳,水蛭组、大黄组抑制作用较强,优于黄芪组。结论:证实了化瘀、泄浊中药水蛭、大黄在抑制MC产生FN及Col-Ⅳ方面优于益气类中药黄芪,复方+雷公藤组作用效果最佳。Objective:To research the effect of Huangqi, Shuizhi, Dahuang and their compounds inhibiting rat mesangium cells to produce fibronectin and type IV collagen. Methods : On the basis of rat mcsangial cells culture in vitro, the rat mesangial cells hyperplasia model was induced by LPS into nine groups : control group, model group, matched group, Huangqi group, Shuizhi group, Dahuang group,tripterygium wilfordii group, compounds group, Compound + tripterygium wilfordii group. ELISA method was used to test expressions of FN and Col - IV by MC, which were influenced by Huangqi, Shuizhi, Dahuang and their compounds. Results : Each serum containing medicine group significantly lowered producations of FN and Col - IV by rat MC which were stimulated by LPS. Shuizhi group's inhibition is better than Huangqi group's. Conclusion:It is confirmed Shuizhi and Dahuang were better than Huangqi in inhibiting rat mesangium cells to produce fibronectin and type IV collagen, and the effect of com- pound + tripterygium wilfordii group was the best.
