摘 要:目的探讨IFN7诱导蛋白10(CXCL10)单核苷酸多态性(SNP)位点rs4257674和rs4256246的多态性和中国人群慢性乙型肝炎(CHB)遗传易感性的关系。方法采用多通道Snapshot分型技术检测501例CHB患者(CHB组)和301例自限性乙型肝炎者(对照组)CXCL10基因rs4257674和rs4256246的多态性,计算和分析基因型、等位基因频率和单体型频率。结果CXCL10 rs4257674位点多态性在CHB组和对照组组中的等位基因频率(OR=2.347,95%CI:1.478—3.729,z2=13.763,P〈0.01)和基因型分布(x^2=12.910,P〈0.01),差异有统计学意义;rs4256246位点多态性在CHB组和对照组中的基因型分布(x^2=1.795,P〉O.05)、等位基因频率(OR=1.107,95%CI:0.897~1.366,x^2=0.899,P〉0.05)差异无统计学意义。rs4257674和rs4256246两位点构成的单体型C/I在CHB组和对照组频率分布差异有统计学意义(OR=2.139,95%CI:1.340—3.413,x^2=10.597,P〈0.01);单体型C/C、T/C频率分布在两组之间差异无统计学意义(x^2C/C=0.262,x^2T/C=1.342,P〉0.05);单体型T/T频率〈0.03。结论CXCL10基因多态性位点rs4257674可能与中国人群CHB发病相关。Objective To investigate the association between two SNPs(rs4257674, rs4256246) in CXCL10 and the genetic susceptibility of Chinese people to chronic hepatitis B infection. Methods A case-control study of 501 Chinese cases of chronic hepatitis B(CHB) was perfonmed and 301 self-limiting HBV-infected individuals were selected as controls. SNPs in CXCL10(rs4257674, rs4256246)were genotyped with the Multiplex Snapshot technique. The genotype, allele and haplotype frequencies were calculated and analyzed. Results The SNP site rs4257674 showed a significant association with the two groups both in allelic analysis( OR = 2.347,95% CI:1.478-3.729, x^2= 13.763, P 〈 0.01) and in genotype analysis(x^2 = 12.910, P 〈 0.01). The other SNP site rs4256246 was not related with two groups both in allelic analysis( OR = 1.107,95% CI:0.897-1.366, x^2 = 1.795, P 〉 0.05) and in genotype analysis( x^2 = 0.899, P 〉 0.05). C/T haplotype, generated by the two SNPs showed significantly difference between the two groups ( OR = 2.139, 95% CI:1.340-3.413, Z2= 10.597, P 〈 0.01). C/C and T/C haplotypes showed no assosition between the two groups (x^2 = 0.262, x^2 = 1.342, P 〉 0.05). T/T haplotype frequency was less than 0.03. Conclusions The data suggests that the SNP site rs4257674 of CXCL10 may be associated with HBV infection in the Chinese population.
