摘 要:目的本文旨在探讨母子分离应激对complexinⅡ(Cplx2)基因小鼠中枢神经递质多巴胺分泌的影响。方法实验分4组:应激Cplx2-/-组、应激Cplx2+/+组、无应激Cplx2-/-组和无应激Cplx2+/+组。无应激组小鼠正常饲养,应激组小鼠从出生后第2日至第21日每天施加母子分离应激。小鼠生长到第4周时,利用聚合酶链反应技术检测其基因型。4组小鼠腹腔注射激动剂去氧麻黄碱或生理盐水后,采用高效液相加电化学检测器测定纹状体多巴胺含量。结果应激Cplx2-/-组、应激Cplx2+/+组、无应激Cplx2-/-组和无应激Cplx2+/+组注射去氧麻黄碱后,与生理盐水对照组相比,纹状体多巴胺水平显著升高(P<0.05),以应激Cplx2-/-组纹状体多巴胺分泌增多最为明显。与无应激Cplx2+/+组相比较,应激Cplx2+/+组注射去氧麻黄碱后纹状体多巴胺分泌增多(P<0.05);与无应激Cplx2-/-组相比较,应激Cplx2-/-组注射去氧麻黄碱后纹状体多巴胺分泌增多(P<0.05);与应激Cplx2+/+组相比,应激Cplx2-/-组注射去氧麻黄碱后纹状体多巴胺分泌增多(P<0.05)。与无应激Cplx2+/+组相比,无应激Cplx2-/-组注射去氧麻黄碱后纹状体多巴胺分泌增多(P<0.05)。结论本实验证实小鼠中枢神经递质多巴胺分泌可能受去母子分离应激和Cplx2等因素的多重影响。【Objective】 To investigate the influence of neonatal isolation stress on secretion of dopamine in CNS of complexin Ⅱ(Cplx2) knockout mouse.【Methods】 The mice were randomly divided into 4 groups: Cplx2-/-with stress,Cplx2+/+ with stress,Cplx2-/-without stress and Cplx2+/+ without stress.Isolation stress was employed on the pups of stress groups from the 2nd day after the postnatal to the 21st day.The PCR was used to determine the gene type and High Performance Liquid Chromatography(HPLC) was employed to detect secretion of dopamine in striatum of CNS.【Results】 The animals of all groups increased secretion of dopamine after injection of 0.2 mg/kg methamphetamine in different levels(P〈0.05).The Cplx2-/-/Cplx2+/+ with stress mouse revealed a more increase secretion of dopamine after injection of methamphetamine compared with Cplx2-/-/Cplx2+/+ without stress mouse(P〈0.05).Indicating is that stress has effect on the secretion of dopamine.When Cplx2-/-mouse with stress/without stress compared with Cplx2+/+mouse with stress/without stress,Cplx2-/-mouse revealed more secretion of dopamine after injection of methamphetamine(P〈0.05),indicating that Cplx2 has effect on the secretion of dopamine as well.【Conclusion】 These results suggest an involvement of stress and Cplx2 in secretion of dopamine in striatum of CNS.
