摘  要:通过数理统计的方法优化双歧杆菌产生物膜培养基。试验首先采用Plackett-Burman试验确定NaCl、MnSO4和半乳糖为影响双歧杆菌生物膜生成的主要因素,再利用正交试验和中心组合试验确定最佳浓度。结果表明:MnSO4、半乳糖和NaCl的最佳浓度分别为0.005g/L,25g/L和0.1mol/L。The medium for biofilm formation by B. bifidium was optimized by statistical methodology for the first time. Two-level factorials of Plackett-Burman design was applied to evaluate the influence of related nutrients on biofilm formation. The sodium chloride, manganese sulfate and galactose were confirmed as critical factors in the medium. The orthogonal design was used to determine the optimal region of these factors for biofilm formation. Then the optimized level of these factors was deter-mined by response surface methodology. The predicted maximal biofilm formation appeared at the region where the concentrations of manganese sulfate, galaetose and sodium chloride were 0. 005 g/L, 25 g/L and 0.1 mol/L, respectively. The correlation between predicted value and measured value of these experiments proved the validity of the response model. The biofilm formation increased 1.5 times in the optimized medium compared to that in the original medium.
