重症肌无力患者胸腺提取液电泳及其活性分析

摘　　要：目的 :深入探讨胸腺在重症肌无力 (MG)发病中的作用。方法 :采用SDS PAGE电泳对MG患者胸腺提取液进行分析 ,并用MTT法研究了胸腺提取液对外周血单个核细胞和胸腺细胞增殖的影响。结果 :30 %MG患者胸腺提取液中存在异常蛋白 ,其相对分子质量小于 144 0 0 ,而在先天性心脏病患者胸腺细胞培养上清中未见这种蛋白。异常蛋白组MG患者的胸腺提取液对正常人外周血单个核细胞 (PBMC)增殖有显著的抑制作用 ,与非自身免疫病患者相比 ,差异有显著性 (P <0 .0 1) ,与其他非异常蛋白组MG患者相比差异有显著性 (P <0 .0 5 ) ,但对胸腺细胞的增殖未见显著差异。结论 :MG患者免疫功能的异常与胸腺提取液中小分子异常蛋白的存在有关 ,与MG的发病有关。Aim: To investigate the role of thymus of patients with myasthenia gravis(MG). Methods: The thymus extract was prepared from thymus of patients with MG, the protein was assayed with SDS polyacrylamide gel electrophoresis (SDS PAGE). Effects of thymus extract on the proliferation of thymocytes and peripheral blood mononuclear cell(PBMC) were evaluated with MTT assay. Results: There was abnormal protein, which molecular weight was less than 14 400 in the thymus extract of 30% patients with MG. The inhibition of thymus extract containing abnormal protein on proliferation of PHA active normal human PBMC was obviously increased, and compared with that of other patients with MG, there was remarkable difference( P <0 05).Conclusion: The thymus extract of patients with MG possesses obvious abnormality, the abnormal protein in thymus extract of patients with MG is accociated with etiology of MG.
