摘 要:目的 探讨阿霉素磁性蛋白微球联合外磁场体内外抑制肿瘤生长的机制。方法 Wistar大白鼠 3 2只 ,用MTT法检测阿霉素磁性蛋白微球对Walker 2 5 6细胞的抑制率及半数抑制剂量 ;观察其体内靶向治疗敏感细胞鼠种植性胃肿瘤的疗效。结果 阿霉素磁性蛋白微球与游离阿霉素体外对肿瘤细胞的生长抑制率相似 (P >0 .0 5 ) ,联合外磁场后其抑制率作用明显增强 (P <0 .0 1)。靶向组对种植瘤的抑制率达 82 .5 2 % (P <0 .0 1) ,荷瘤动物生成时间明显延长 ,延长率达 2 86.3 8% (P <0 .0 1)。结论 阿霉素磁性蛋白微球体内外仍具有抑制敏感肿瘤细胞生长的能力 ,联合磁场可发挥靶向、高效的抗瘤作用。Objective To study the mechanism of adriamycin magnetic albumin microspheres (ADM MAMs) inhibiting the growth of the malignant tumor cells in vitro and in vivo. Method The growth inhibitory rate (IR) and 50 % inhibitory concentration (IC50) of ADM MAMs to Walker 256 cells were assayed by MTT colorimetric method. The therapeutic effects were evaluated about the ADM MAMs combination with external magnetic field against the implanted gastric tumors reconstructed by the sensitive tumor cells in rats. Results The IR and IC50 of the ADM MAMs against the tumor cells were similar to those of the free adriamyic (ADM) (P>0.05), obviously increased by addition of the external magnetic field (P<0.01) in vitro. In vivo, the growth inhibition of the implanted gastric tumor in the targeting therapy group was 82.52 % and the survival of the animals with the implanted tumor in the targeting therapy group was obviously prolonged with the prolonged rate being 286.38 % (P<0.01). Conclusion The ADM MAMs could inhibit the growth of the sensitive tumor cells in vitro and in vivo. ADM MAMS combined with external magnetic field showed targeting and high effective antitumor activities.
