摘 要:目的从黄精中分离纯化低聚糖样品并进行结构分析。方法采用80%乙醇回流提取,D101大孔吸附树脂柱层析、活性炭柱层析和Sephadex LH-20凝胶柱层析分离纯化,得到黄精低聚糖样品。采用化学分析方法研究黄精低聚糖样品的理化性质,并通过高效凝胶渗透色谱(HPGPC)、气相色谱(GC)、紫外光谱(UV)、红外光谱(IR)、核磁共振氢谱(1HNMR)和核磁共振碳谱(13C NMR)等方法对黄精低聚糖样品进行结构分析。结果黄精低聚糖样品纯度较高,为均一性组分,平均分子量为1 471 Da,由单一葡萄糖组成的D-吡喃葡聚糖,分子结构中葡萄糖的连接方式主要为1→3和1→4,苷键构型以β-糖苷键为主,还含有少量的α-糖苷键。结论首次从黄精中分离并鉴定了黄精低聚糖。AIM To study the isolation, purification and characterization of Polygonati Rhizoma oligosaccha- ride (PRO). METHODS Polygonati Rhizoma was extracted by 80% ethanol, and further isolated and purified by D101 macroporous adsorption resin column chromatogramphy, activated carbon column chromatography and Sephadex LH-20 gel column chromatography. The structural analysis was identified by high performance gel perme-ation chromatography (HPGPC), gas chromatography (GC), ultraviolet spectrometry (UV), infared spectroscopy ( IR), NMRspectroscopy ( ^1 H, ^13 C) . RESULTS PRO was a single homogeneous component and the average molecular weight was estimated at 1 471 Da. The D-pyran glucan consisted entirely of glucose units, which were linked in straight chains by 1→4 bond and 1→6 bond, most of the constituent gluc0sidic bond were jointed inβ- form with a little of α-form. CONCLUSION The PRO is obtained from Polygonati Rhizoma for the first time.
