布鲁菌PCR检测方法的建立及其应用

摘　　要：本文根据已发表布鲁菌BCSP31基因序列,设计1对引物,以布鲁菌19号苗基因组DNA作为模板,建立诊断牛布鲁菌病的PCR方法,并对方法的特异性、敏感性及适用性进行验证。结果显示,PCR能扩增布鲁菌BCSP31基因特定序列。该方法对布鲁菌的最小检出量为9pg,对葡萄球菌26001株、大肠杆菌O78等7种参照菌株的核酸扩增结果均为阴性。建立的PCR方法具有快速简便、特异性强、敏感性高等特点。In this research,one pair of primers were designed according to the sequence of Brucella genus gene BCSP31 reported,and the specificity,sensitivity and applicability of this method were verified.Results showed that Brucella genus gene BCSP31 specific DNA fragment could be detect by PCR established in this research.The PCR assay could detect as low as 9 pg of Brucella 19 vaccine strain DNA.All the detection results of seven Brucella species by this method were negative.Results indicate that a rapid,specific and sensitive PCR method for detection of Brucella was developed,and can be used for brucellosis diagnosis in clinic.
