摘  要:目的 观察大鼠角膜新生血管(CNV)形成过程中环氧酶-2(COX-2)和VEGF的表达,探讨结膜下注射环氧酶-2 抑制剂尼美舒利对角膜CNV的抑制作用.方法 HE染色分别观察和计算CNV面积,免疫组织化学方法检测碱烧伤后COX-2,VEGF在角膜中的分布,比较处理组和对照组COX-2,VEGF的表达量,并计算两者的相关性.结果 Ⅰ组和Ⅴ组CNV面积明显高于其它各组(P〈0.05).第4和第7天时,Ⅱ组和Ⅳ组CNV面积明显低于其它各组(P〈0.05).第4天时,Ⅳ组CNV面积明显低于Ⅱ组(P〈0.05).碱烧伤后第7及14天,Ⅱ~Ⅳ组COX-2,VEGF表达量远低于第Ⅴ组.结论 COX-2在角膜CNV形成中表达增加,通过调控VEGF的表达而起重要作用,尼美舒利抑制COX-2的表达,抑制CNV形成.Objective To evaluate the inhibitory effect of subconjunctival injection of nimesulide on rat corneal neovascularization ( CNV ) and the expression vascular endothelial growth factor(VEGF) and COX-2. Methods The growth of CNV was observed every day by slit lame corneal microscope and neovascularization areas calculated. Expression of COX-2 and VEGF were examined by immunohistochemistry,on day 2,4,7and 14 after operation in the cornea. Results The CNV grew quickly in 2 - 4 days and reached peak in the 7th day after corneal cautery. The mean areas and expression of COX-2 and VEGF of CNV in group II and IV was sifnificantly smaller than that of other groups at the 4 and 7day ( P 〈 0.05 ) , but no significant differernce was found between group I and V ( P 〉 0.05 ) at any time of the ex- periment. Conclusion The activity of COX-2 grow significantly in the stage of CNV. Topically administration of moderate nimesulide has sig- nificant effects on the inhibition of CNV, through descending the expression of VEGF.
