摘  要:目的研究RNA干扰沉默S100A2和E-cadherin基因后对胃癌细胞增殖和侵袭力的影响。方法采用RNA干扰技术,在E-cadherin和S100A2表达阳性的MNK45细胞同时沉默E-cadherin和S100A2基因表达,逆转录聚合酶链反应和Western blot检测干涉效果,MTT和Transwell检测细胞生长和侵袭力的变化。结果小干扰RNA明显抑制MKN45细胞E-cadherin和S100A2的表达;干涉第11天,E-cadherin和S100A2蛋白表达下降,细胞生长速度和细胞侵袭力明显增加。结论 E-cadherin和S100A2蛋白具有抑制细胞增殖和侵袭的功能,可作为抑癌因子参与胃癌的发生与转移。Objective To study the effects of gene silencing in S100A2 and E-cadherin by RNA interference on gastric cancer cell proliferation and invasiveness.Methods The expression of E-cadherin and S100A2 genes was silenced by RNA interference in the cell line MKN-45 in which E-cadherin and S100A2 genes are positively expressed.The effect of RNA interference was detected by RT-PCR and Western blot,and the changes of cancer cell proliferation and invasiveness were detected by MTT and Transwell.The effects of E-cadherin and S100A2 genes on gastric cancer were finally analyzed by RNA interference in the study.Results The results showed that the synthesis of E-cadherin and S100A2 mRNA was suppressed dramatically after 7 days triggered by small interfering RNA.On the 11th day of interference,the expression of E-cadherin and S100A2 protein was lower.MTT assay was performed to observe the cell growth rate.Conclusion E-cadherin and S100A2 work as cancer suppressor genes in the tumorogenesis and metastasis of gastric cancer with the function of inhibiting the proliferation and invasiveness of gastric cancer cell.
