摘 要:目的了解普伐他汀对2型糖尿病大鼠视网膜组织中转化生长因子β1(TGF-β1)表达的影响,探讨普伐他汀对糖尿病性视网膜病变的防治作用。方法将54只Wistar大鼠随机分为正常组、糖尿病组和普伐他汀治疗组,每组18只。用链脲佐菌素诱导糖尿病组和普伐他汀治疗组大鼠建立2型糖尿病模型,正常组和糖尿病组正常饮食,普伐他汀治疗组在正常饮食中加入普伐他汀(100 mg.kg-.1d-1)。30周后取各组大鼠视网膜,分别用免疫组化法和Real-time PCR法检测视网膜组织中TGF-β1的表达水平。结果 TGF-β1阳性表达于视网膜神经节细胞胞质中。糖尿病组视网膜组织变薄,神经节细胞数减少;与正常组相比,普伐他汀治疗组视网膜形态无明显改变,神经节细胞数无明显减少。糖尿病组视网膜组织中TGF-β1表达明显高于正常组(P<0.05),普伐他汀治疗组明显低于糖尿病组(P<0.05)。结论普伐他汀能够阻止或延缓糖尿病视网膜形态改变,能够降低糖尿病大鼠视网膜组织中TGF-β1的表达水平。Objective To investigate the effects of pravastatin on the expression of TGF-β1 in the retina of type Ⅱ diabetes rats,and the preventive effect of pravastatin in the pathological changes in diabetic retina.Methods Fifty-four Wistar rats were randomly divided into three groups:normal group,diabetic group and pravastatin treatment group,with 18 rats each.Rats in diabetic group and pravastatin treatment group were induced with streptozotocin to type Ⅱ diabetes rat model.The rats in the first two groups were fed with normal diet and the rats in the pravastatin treatment group were fed with normal diet plus pravastatin 100 mg·kg-1·d-1.After 30 weeks,rats in all groups were sacrificed.The expression of TGF-β1 in retinal tissue was detected with immunohistochemistry and Real-time PCR.Results Positive expression of TGF-β1 was in cytoplasm of retina ganglion cells.Compared with the normal group,the thickness of retina became thinning and the number of ganglion cells decreased in the diabetic group,while those were not significantly changed in the pravastatin treatment group.The expression level of TGF-β1 in diabetic group was significantly higher than that in the normal group(P 〈 0.05),and the expression level of TGF-β1 in pravastatin treatment group was significantly lower than that in diabetic group(P 〈 0.05).Conclusion Pravastatin could control and delay the morphological changes of diabetic retinopathy,and pravastatin could decrease the expression of TGF-β1 in diabetic retina.
