摘 要:目的研究肝癌组织和人肝癌细胞系中甘油二酯激酶ζ(diacylglycerol kinase,DGKζ)和蛋白激酶C(PKC)的表达及意义。方法应用免疫组织化学和细胞化学法检测DGKζ和PKC在60例肝癌与癌旁组织、10例正常肝组织和体外培养的正常人肝细胞系、癌旁肝细胞系和人肝癌细胞系的表达情况,对肝癌组织中DGKζ和PKC的表达进行相关性分析。结果 DGKζ在正常肝组织呈阴性表达,在癌旁组织肝细胞胞质和肝癌细胞的细胞膜上有阳性表达;肝癌组织中DGKζ的阳性率为86.7%(52/60),显著高于癌旁组织(40/60,χ2=6.708,P=0.010)和正常肝组织(阴性,χ2=33.704,P<0.000 1)。PKC表达在正常肝细胞的胞质和胞核、癌旁组织肝细胞的细胞膜和肝癌组织的胞质或胞膜;肝癌组织中PKC的阳性率为71.7%(43/60),与癌旁组织61.7%(37/60,χ2=1.350,P=0.245)和正常组织100.0%(10/10,χ2=3.742,P=0.104)相比无统计学差异,癌旁组织中PKC的阳性率显著低于正常肝组织(χ2=5.709,P=0.025)。肝癌组织中DGKζ与PKC的表达呈正相关(r=0.296,P=0.022);DGKζ和PKC均主要表达在三种细胞系肝细胞的胞质。结论 DGKζ主要在肝癌组织被激活,可能有抑制肝癌细胞增殖的作用。Objective To investigate the expression and significance of diacylglycerol kinase ζ(DGKζ)and protein kinase C(PKC) in human hepatocellular carcinoma(HCC) and hepatocellular carcinoma cell lines. Methods Immunochemistry was employed to detect the expression of DGKζ and PKC in 60 cases of HCC,adjacent liver tissues,10 normal liver tissues,normal liver cell line,HCC cell line and their adjacent liver cell line.The relationship between DGKζ and PKC expression in HCC was analyzed. Results DGKζ was positive in hepatocyte cytoplasm of tumor adjacent tissues and cell membrane of HCC tissues,but negative in normal liver tissues.The positive rate of DGKζ in HCC tissues was significantly higher than those in carcinoma adjacent tissues [86.7%(52/60) vs 66.7%(40/60),χ2=6.708,P=0.010] and normal liver tissues(86.7% vs 0,χ2=33.704,P0.000 1).PKC was positive in hepatocyte cytoplasm and nucleus of normal liver tissues,hepatocyte membrane of carcinoma adjacent tissues and cell cytoplasm or membrane of HCC tissues.The positive rate of PKC in HCC tissues was 71.7%(43/60),and it was not statistically different from those in carcinoma adjacent tissues [61.7%(37/60),χ2=1.350,P=0.245] and normal liver tissues [100.0%(10/10),χ2=3.742,P=0.104].The positive rate of PKC in carcinoma adjacent tissues was significantly lower than that in normal liver tissues(χ2=5.709,P=0.025).There was positive correlation between the expression of DGKζ and PKC(r=0.296,P= 0.022).Both DGKζ and PKC were expressed mainly in cytoplasm of all three cell lines. Conclusion DGKζ is activated mainly in HCC tissues,and may inhibit the proliferation of carcinoma cell.
