摘 要:目的观察miR-518b在乳腺癌中的表达,以及对人乳腺癌细胞株MCF-7增殖、细胞周期、凋亡和侵袭能力的影响。方法运用定量逆转录聚合酶链反应(PCR)检测乳腺癌及正常组织中miR-518b的表达量,结合临床病理资料,分析其异常表达与各病理因素的相关性。并运用噻唑蓝(MTr)比色法了解其增殖,流式细胞仪分析细胞的周期、凋亡,Transwell实验观察其对细胞侵袭能力的影响。结果乳腺癌组织中miR-518b表达水平为2.751585±5.856351,正常组织中为7.334841±9.843424,癌组织中miR-518b表达明显低于正常组织(P〈0.01)。miR-518b在乳腺癌中的表达异常与淋巴结转移相关(P〈0.01),与病理分期、肿瘤大小、原癌基因人类表皮生长因子受体2(HER-2)、雌激素受体(ER)、孕激素受体(PR)、pS3无明显相关性。在48h浓度为150nmoVL时,miR-518b对乳腺癌细胞的抑制作用最明显,抑制率为59.9%。与空对照组和负对照组比较,G0/G1期细胞未见明显增多(75.52±1.78)%,S期无明显减少(9.52±0.91)%,早期凋亡细胞比例未见明显升高(2.60±0.06)%(P〉0.05)。Transwell实验结果显示随着miR-518bmimics浓度的增加,穿出小室膜肿瘤细胞大致呈递减趋势,并与对照组差异明显(P〈0.01)。结论miR-518b在乳腺癌患者中异常表达,癌组织中的表达明显低于正常组织,miR-518b对乳腺癌细胞株的增殖和侵袭能力有明显抑制作用,其在肿瘤发生发展中可能担任着抑癌基因的角色。Objective To explore the expression of miR-518b in invasive ductal breast cancer and analyze its impact on the proliferation, cell cycle, apoptosis and invasion of MCF-7 breast cancer cell line. Methods Twenty pairs of breast cancer and normal tissue specimens were chosen after confirmation of breast invasive ductal carcinoma. We conducted reverse transcription-polymerase chain reaction (RT-PCR) to quantify the expression of miR-518b, and its correlation with clinical data was analyzed by using SPSS software 13.0. To characterize the effect of miR-518b on MCF-7 breast cancer cells, functional assays were respectively employed, including proliferation, cell cycle, apoptosis, and Transwell assays. Results The relative expression of miR-518b in breast cancer specimens was 2. 751 585±5. 856 351, which was significantly lower than 7. 334 841±9. 843 424 of adjacent normal ones ( P 〈 0.01 ). Aberrant expression of miR-518b was correlated with lymphatic metastasis (P 〈 0.01 ). On the contrary, no correlation was observed of miR-518b with TNM stage, tumor size, HER-2, estrogen receptor (ER), progesterone receptor (PR) and 1353 status. Methyl thiazol tetrazolium (MTr) assay revealed that miR-518b mimics significantly inhibited proliferation of breast tumor cells at 48 h after transfection at the concentration of 150 nmol/L with the inhibition rate being 59.9%. In comparison to controls, there was no significant difference in the percentage of G0/G1 phase cells ( 75.52±1.78 ) % , S phase cells ( 9. 52±0. 91 ) % and the proportion of early apoptotic cells ( 2. 60±0. 06) % after miR-518b mimics transfection ( P 〉 0.05 ). The amount of invaded breast cancer cells in miR-518b transfected groups was significantly different from control groups and declined with an increased miR-518b concentration. Conclusion miR-518b expression is aberrantly expressed in breast cancer. Breast tumor specimens show decreased miR-518b expression compared to their normal counterparts. Considering the
