摘  要:目的:采用离子交换层析法分别从海地瓜和冰岛刺参中,分离纯化两种海参岩藻聚糖硫酸酯(SC-FUC),通过建立小鼠Lewis肺癌自发性肺转移模型,比较分析两种SC-FUC对小鼠肿瘤生长和转移的抑制作用。方法:采用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生高效液相色谱法(HPLC)测定其单糖组成,采用离子色谱法测定其硫酸基含量。通过连续腹腔注射两种SC-FUC 21d,剥离原位肿瘤和双侧肺,分别称瘤质量和计数肺表面转移灶结节数。采用RT-PCR法检测肿瘤组织中缺氧诱导因子(HIF-1α)和血管内皮生长因子(VEGF)mRNA的表达。结果:海地瓜SC-FUC和冰岛刺参SC-FUC均由岩藻糖组成,硫酸基含量分别为26.3%和29.31%;两者均能显著抑制荷瘤小鼠原位肿瘤的生长,抑瘤率分别为19.24%和28.69%;显著减少肺表面转移灶结节数(P<0.01),转移抑制率分别为31.4%和57.1%;显著降低荷瘤小鼠肿瘤组织中HIF-1α和VEGF mRNA的表达(P<0.05,P<0.01)。冰岛刺参SC-FUC的效果要优于海地瓜SC-FUC。结论:两种SC-FUC均能显著抑制肿瘤的生长和转移,其作用机制与其高硫酸化程度有关。Purpose: To investigate the influence of fucoidan (SC-FUC) from Cucumariafrondosa and Acaudina molpadioidea on the growth and metastasis of Lewis lung carcinoma in mice. Methods: Lewis lung carcinoma models of C57BL/6J mice were established. SC-FUC was administrated to mice by intraperitoneal injection for 21 consecutive days. Mice were then sacrificed and the subcutaneous tumors were weighed to calculate inhibitory rate. The metastatic tumor foci on lung surface were counted to calculate occurrence rate and inhibitory rate of metastases. The expression of H1F-lα and VEGF mRNA was detected by RT- PCR. Results: Tumor growth was suppressed significantly in SC-FUC groups (P〈0.05), the inhibition rates were 19.24% and 28.69% for SC-FUC from Cucumaria frondosa and Acaudina molpadioidea, respectively. The metastatic foci occurrence rates ofSC FUC groups were lower than that of control group and the differences were significant (P〈0.05). SC-FUC significantly down-regulated the expression of H1F-1α and VEGF mRNA. The inhibitory effect of SC-FUC from Cucumariafrondosa was greater than that of SC-FUC from Acaudina molpadioidea. Conclusions: The two SC-FUC from Cucumariafrondosa and Acaudina molpadioidea, respectively could inhibit the growth and metastasis of Lewis lung carcinoma in mice. The mechanism of inhibition may be related to the sulfation of SC-FUC.
