摘 要:目的 :观察蝎毒抗癌多肽 (APBMV)对肝肿瘤的抑制作用。方法 :噻唑蓝 (MTT)法、台盼蓝拒染法、集落形成以及H2 2 荷瘤小鼠模型。结果 :APBMV导致SMMC - 772 1细胞代谢MTT的能力显著低于对照组 ,APBMV的IC50 为 11 33μg/mL ;APBMV能显著抑制该细胞的生长 ,剂量 -效应关系明显 ,其IC50 分别为 15 87μg/mL、13 0 5 μg/mL和 8 70 μg/mL ;当APBMV的浓度 >8μg/mL时 ,SMMC - 772 1细胞的集落形成率显著低于对照组。APBMV能明显抑制H2 2 带瘤小鼠肿瘤的生长 ,生长抑制率达 37 31% (P <0 0 1) ,H2 2 带瘤小鼠外周血白细胞计数和脾脏指数无明显变化或略高于对照组 ,而 5 -Fu治疗组H2 2 带瘤小鼠白细胞计数和脾脏指数均明显下降(P <0 0 1)。结论 :APBMV是存在于东亚钳蝎蝎毒中的一种有效低毒的抗肿瘤成分 ,能够抑制SMMC - 772 1和H2 2 两种肝癌细胞的生长。AIM: To observe the growth inhibiting effect of antineoplastic polypeptide from Buthus Martensii venom(APBMV)on liver neoplasm METHODS: MTT calorimetric method, trypin blue exclusion,colony formation and H 22 -bearing mouse model RESULTS: The ability to metabolize MTT of SMMC-7721 cells was lower than control distinctly after the cells were treated by APBMV The IC 50 of APBMV was 11 3 μg/mL The growth of SMMC-7721 cells was inhibited obviously by APBMV and the dose-response relationship was clear, the IC 50 were 15 87 μg/mL,13 05 μg/mL and 8 70 μg/mL respectively The colony formation rate of SMMC-7721 cells was also decreased evidently compared with control when treated concentrations of APBMV were higher than 8 μg/mL Tumor growth of H 22 -bearing mice was inhibited by APBMV evidently, the growth inhibiting rate was reached 37 31%( P <0 01) After treatment by APBMV, white blood cell(WBC) number and spleen index of H 22 -bearing mice were not changed markedly or slightly higher than control group In 5-fluorouracil treated group,WBC number and spleen index of H 22 bearing mice were all reduced distinctly( P <0 01) CONCLUSION: APBMV was one kind of effective and low toxicity antineoplastic components in the venom of Buthus martensii scorpion It possesses growth inhibiting ability on SMMC-7721 and H 22 hepatoma cells
