摘  要:目的探讨Ucn3在肠易激综合征(irritable bowel syndrome,IBS)大鼠模型下丘脑中的表达方式。方法采用孤养附加慢性不可预见性刺激建立IBS模型,用体质量变化、排便粒数、敞箱行为评分和蔗糖水偏嗜度评价动物模型;建成后留取大鼠下丘脑,采用免疫组化法及RT-PCR法检测各组大鼠下丘脑中Ucn3表达水平的变化。结果正常对照组、模型组体质量评分为(392.93±21.75)分和(331.56±25.04)分;粪粒数为(2.71±2.36)粒和(9.54±3.92)粒;敞箱实验评分为(81.39±10.87)分和(17.92±8.03)分;蔗糖水偏嗜度评分为(81.39±10.87)分和(68.59±19.51)分。免疫组化法检测Ucn3阳性表达率为(14.32±0.54)%和(18.34±0.85)%。RT-PCR法测定Ucn3的表达为(8.58±3.06)和(19.83±2.95)。模型组与对照组相比,Ucn3的表达升高,经one-way ANOVA及SNK-q检验,差异有统计学意义(P<0.05)。结论 Ucn3在IBS大鼠模型下丘脑中表达升高,为进一步明确IBS的发病机制提供了新的证据。Objective To explore the expression of Ucn3 in hypothalamus of irritable bowel syndrome (IBS) rat model. Methods The IBS models were established by isolated feeding and additional chronic unpredictable stimulus. The models were assessed by weight, counting the feces granules, open-field behavior score and fluid consumption test. After that, rat hypothala- muses were used to detect the Uen3 expression level with the immunohistochemistry and real time (RT)- PCR method. Results In the normal control group and model group, the weight scores were 392.93±21.75 and 331.56±25.04;the feces granules were 2.71±2.36 and 9.54±3.92 ;the open-field behavior scores were 81.39±10.87 and 17.92±8.03 ;the scores of the fluid consumption test were 81.39±10.87 and 68.59±19.51, respectively. The positive expression rates of Uen3 with immunohistoehemistry were (14.32±0.54)% and (18.34±0.85)% respectively. The expressions of Ucn3 by RT-PCR method were (8.58±3.06) and (19.83±2.95 ) respectively. Compared with the normal control group, the weight, counting the feces granules ,open-field behavior scores and fluid consumption test of the model group were significantly improved (P〈0.05) and the expression of Uen3 in model group was increased significantly (P〈0.05). Conclusion The expression of Ucn3 is increased in the hypothalamuses of IBS rat model, which provides a new evidence for further research of the pathogenesis of IBS.
