膀胱移行细胞癌人乳头瘤病毒的分子生物学研究

摘　　要：目的　研究人类乳头瘤病毒 6、11、16、18型及P53与膀胱移行细胞癌的关系。方法　采用PCR方法检测了 75例膀胱移行细胞癌组织中人类乳头瘤病毒的感染和P53蛋白表达情况。结果　膀胱移行细胞癌组织 6、11、16、18阳性率分别为 6 .7%、5.3%、33.3%和 6 .7% ,低危型人类乳头瘤病毒、6、11)阳性率为 9.3% (7/ 75) ,高危型人类乳头瘤病毒 (16、18)阳性率为 34.7% (2 6 / 75)。同一膀胱癌组织中两种以上(包括两种 )人类乳头瘤病毒亚型感染 8例 ,占 10 .6 %。人类乳头瘤病毒 6、16、18型感染阳性率在肿瘤有无转移组中差异显著 (P <0 .0 5)、人类乳头瘤病毒 16、18阳性率在肿瘤病理分级中有极显著性差异。人类乳头瘤病毒DNA型别的分布在膀胱癌中以 16型为主。P53蛋白阳性表达率为 4 4 %。结论　高危型人类乳头瘤病毒感染与P53异常表达可能与膀胱癌发生有关。Objective To study the relationship of bladder transitional cell carcinoma with human papillomavirus (HPV) 6、11、16、18 types and P53.Methods We used polymerase chain reaction(PCR) to test HPV 6、11、16、18 types infection from 75 cases with bladder cancer,and usedmununobiochemisty method to delecte p53 expression.Results The positive rales of HPV6、11、16、18types were 6.7%、5.3%、33.3% and 6.7% respectively.The positive rate of low risk types HPV(16/18) Was 9.3%(7/75),,and the that of high risk types HPV(16/18) was 9.3%(7/75),and the that of high risk types HPV(16/18) was 34.7%(26/75).There were 8 cases of two HPV type mixed infection s m the same sampies from 75 cases with blader cancer tissue.the positive rates of HPV 6、16、18types in tumor metastasis group were significantly higher than that of non metastasis group.The positive rates of HPV 16、18types in G3 were significantly higher than that of G1 and G2.HPV 16 type are the main type in bladder cancer.The Positive rate of P53 expression was 44%.Conclusion These results implicated that the occurrence and development of bladder transitional cell carcinoma may be closely related to the high risk types HPVs infection and P53 expression.
