摘  要:目的:探讨不同淋巴细胞分离液对小鼠骨髓单个核细胞生物学活性的影响。方法:使用Ficoll、Percoll和小鼠淋巴细胞分离液分离小鼠骨髓单个核细胞,分别在显微镜下观察细胞形态,胎盘蓝染色法计数细胞数量并检测细胞活力,MTT法检测细胞增殖能力,采用流式细胞仪检测细胞周期,并分别计数三种淋巴细胞分离液分离出的小鼠骨髓单个核细胞,比较不同分离液对小鼠骨髓单个核细胞分离率的影响。结果:使用小鼠淋巴细胞分离液及Percoll分离液获得的小鼠骨髓单个核细胞较Ficoll分离出的细胞数量多(P<0.05),但是细胞形态、活细胞率、细胞增殖能力和细胞周期三者均无明显差别(P>0.05)。结论:不同淋巴细胞分离液对小鼠骨髓单个核细胞生物学活性虽无明显差别,但小鼠淋巴细胞分离液使用方便,获取的小鼠骨髓单个核细胞的数量较多。Objective:To explore the biological activity of mouse bone mononuclear ceil which isolated by different lymphocytes separation medium. Methods:Mouse bone mononuclear cell were isolated by Ficoll,Percol and mouse lymphocytes separation medium. Cell morphological structure under light microscope were observed. Cell viability by trypan blue test, proliferation of obtained mouse bone mononuclear cell by MTT, ceil cycle by flow cytometry were detected. Mononuclear cell separation rate which isolated by different lymphocytes separation medium was compared. Results: The use of mouse lymphocytes separation medium and Percoll obtained more bone marrow mononuclear cells compared with Ficoll ( P〈 0.05), but there is no obvious difference in the cell morphology structure, cell survival rate, cell proliferation and cell cycle of three( P ~0.05). Conclusion: there is no obvious difference in the biological activity of mouse bone mononuclear cell which isolated by different lympbocytes separation medium, but mouse lymphocytes separation medium had the advantages of convenient use and got more number of mouse bone marrow mononuclear cells than Ficoll.
