油脂中黄曲霉毒素B_1快速检测试剂盒研究

摘　　要：在胶体金免疫层析技术的基础上,建立了一种快速检测油脂中黄曲霉毒素B1(AFB1)的新方法.本研究的AFB1快速检测试剂盒突破了常规胶体金免疫层析检测试纸只能检测水溶液样品的限制,可对含油脂样品直接检测,工作时无需复杂的油脂样品处理工艺,缩短了预处理时间.其基本原理是利用吐温(Tween 20)对油脂样品进行乳化处理,使油脂在油脂处理液(甲醇-吐温-磷酸缓冲溶液)中均匀分散为微小油滴,形成稳定的水包油型乳化体系,从而使水相中的抗原、抗体反应不受油脂干扰.测试结果表明本试剂盒对油脂中的AFB1的检测限为10ng.mL-1,检测时间为10min,无假阳性、假阴性.A new method based on immunogold labeling was developed for AFB1 detection in oil. This rapid detection kit could be used for AFB1 testing from samples containing fats, oils without complicated sample preparation process. This is a huge advantage over traditional immunogold kits for AFB1 detection, which could be used only to detect samples without lipid or oil. Tween 20 was used to emulsify lipid in water, to construct water-oil emulsion system, this would avoid antigen/antibody reactions in oil droplets on NC membrane. This novel AFB1 kit has a detection limit of 10 ng. mL^-1 , a test time of 10 min, with no false positives or false negatives.
