摘 要:目的研究麦考酚酸(MPA)对翼状胬肉组织成纤维细胞(PFB)活性的影响。方法体外培养PFB及Tenon囊成纤维细胞(TFB),以丝裂霉素C(MMC)为对照,MTT比色法、5-溴脱氧尿嘧啶(5-bromo-2-deoxyuridine,Brdu)掺入法观察MPA对PFB的生长抑制作用,以流式细胞术等检测MPA对PFB、TFB的细胞毒性作用、细胞周期及凋亡的影响。结果 MPA可显著抑制PFB、TFB的增殖,呈时间、剂量依赖性,且不引起TFB的凋亡或坏死。MPA作用后,PFB的G0/G1期比例增加,S期及G2/M期比例减少。结论 MPA对PFB有显著的抑制作用,但不引起TFB的凋亡或坏死,且主要抑制在PFB的DNA合成期。Objective To investigate whether mycophenolic acid(MPA) exerts antifibrotic effects on pterygium fibroblasts(PFB).Methods TFB and PFB were obtained from tissue explants during strabismus or pterygium surgery and identified with vimentin immunohistochemisty staining.MTT was used to find the optimization concentration of MPA and MMC and detect their inhibitory effect on TFB and PFB.Flow cytometry and PI fluorescence staining was used to show the cytotoxicity of MPA to TFB.Brdu fluorescence staining was used to detect the inhibitory effect of MPA on PFB.Results MPA had significant inhibitory effect on PFB,TFB.No apoptotic effect was found on TFB.In the mycophenolie acid groups,the fraction of PFB in the S-phase and G2/M-phase decreased respectively while that in the G0/G1-phase increased.Conclusion MPA exerted significant inhibitory effect on PFB with no apoptotic effect on TFB.
