糖尿病动脉硬化大鼠模型建立的研究

摘　　要：目的建立糖尿病动脉硬化大鼠模型。方法 6周龄雄性SD大鼠40只,采取简单随机抽样法分为对照组和模型组,每组20只。对照组普通饲料喂养及注射枸橼酸缓冲液,模型组普通饲料喂食4周后连续3 d腹腔注射链脲佐菌素(35 mg/kg),再用高脂高糖饲料喂养8周。检测大鼠血糖、TC、TG、HDL、LDL;处死大鼠,观察主动脉并取病变处制作病理切片镜下观察。结果与对照组相比,模型组大鼠给药后72 h、第10天血糖明显增高(P<0.01);高糖高脂喂食8周后,模型组大鼠血脂4项明显增高(P<0.01)。肉眼观察见主动脉肥大,镜下见具有糖尿病动脉硬化的早期改变,如纤维组织增生、有核细胞增多等。结论小剂量连续注射链脲佐菌素联合高糖高脂饮食,可成功建立大鼠糖尿病动脉硬化模型。Objective To establish rat model of type 2 diabetes atherosclerosis.Methods Forty male SD rats（6 weeks old） were randomly divided into two groups：control group（n=20） and model group（n=20）.The control group was fed with normal diet and injected with citrate buffer.After 4 weeks of normal diet,model group was given streptozotocin（STZ）（35 mg/kg） consecutively by intraperitoneal injection for 3 days,and then fed with high-fat and high-sugar diet for 8 weeks.Blood glucose,total cholesterol（TC）,triglyceride（TG）,high density lipoprotein（HDL） and low density lipoprotein（LDL） were measured.The rats were killed to observe pathological changes of aorta.Results Blood glucose of model group on the 3rd,10th day after consecutive STZ injection was significantly higher than that of the control group（P0.01）.After the rats were fed with high-sugar and high-fat diet for 8 weeks,serum lipids of model group was significantly higher than that of the control group（P0.01）.Aorta hypertrophy could be observed by the naked eye.The early pathological changes of type 2 diabetes atherosclerosis,such as fibroplastic proliferation and increase of nucleated cells,could be seen under the microscope.Conclusion The rat model of type 2 diabetes atherosclerosis could be established successfully by consecutive injection of low dose streptozotocin together with high-sugar and high-fat diet.
