摘  要:目的探讨瘦素(leptin)对人牙周膜细胞增殖和骨向分化的作用。方法采用酶消化法体外培养人牙周膜细胞,用不同浓度梯度的leptin处理细胞,通过四唑盐(MTT)比色试验和碱性磷酸酶活性测试,检测leptin对细胞体外增殖分化的影响。将人牙周膜细胞与羟基磷灰石/磷酸三钙材料作为复合体,植入BALBc免疫缺陷裸鼠,12周后采用组织学和免疫组化法检测裸鼠体内生成物。结果体外实验leptin对人牙周膜细胞增殖有明显抑制作用,但leptin能促进人牙周膜细胞碱性磷酸酶的活性表达。体内试验中leptin可提高牙周膜细胞生成类骨质的能力。结论 leptin抑制牙周膜细胞的增殖活性,但可明显促进牙周膜细胞骨向分化。Objective To investigate the effects of leptin on in vitro proliferation and in vivo osteogenic differentiation of human periodontal ligament cells(hPDLCs).Methods hPDLCs were cultured with enzyme-digestion technique.Different concentration of leptin was added to the hPDLCs.MTT assay and ALP activity were used to evaluate the effects on proliferation.hPDLCs were incubated with hydroxyapatite/tricalcium phosphate(HA/TCP) and then transplanted subcutaneously into the back of immunocompromised mice to examine the in vivo formation of calcification.Results Leptin inhibited the proliferation of hPDLCs in vitro but enhanced the expression of ALP activity of hPDLCs in vivo.Histological examination and immunohistochemical staining showed that leptin-treated hPDLCs generated more bone-like structures.Conclusion Leptin significantly inhibited the proliferation of hPDLCs but enhanced odontogenic/osteogenic differentiation.
