摘 要:目的应用蛋白指纹图谱技术筛查前列腺癌患者血清蛋白质表达谱。建筑血清中的特异性标志物。方法选择前列腺癌患者22例和前列腺增生患者26例以及同期正常对照者25例。用表面增强激光解析离子化飞行时间质谱技术检测各血清样本的血清蛋白指纹图谱,比较血清蛋白指纹图谱的差异。结果前列腺癌组的血清蛋白质谱图中质荷比为3165、3982、4291的蛋白质相对含量(9.794±4.449、3968±2.247、11.020±6.069)高于正常对照组(5062±2.245、1.823±1.432、6.525±5.270),而质荷比为2747、10268的蛋白质相对含量(22.390±12179、0953±O.338)低于正常对照组(42.231±16970、1.932±O.864);质荷比为10268的蛋白质的相对含量前列腺癌组(0.953±0.338)低于前列腺增生组(1.314±0.493)。差异均有统计学意义(均P〈0.05)。但前列腺增生组与正常对照组差异均无统计学意义(均P〉0.05)。PSA阳性的前列腺癌组质荷比为3960的蛋白质相对含量(3.447±1.434)于PSA阳性的前列腺增生组(1.473±O971),而PSA阳性的前列腺癌组质荷比为10268的蛋白质相对含量(O.847±O.408)低于PSA阳性的前列腺增生组(1.538±0.182),差异均有统计学意义(均P〈0.05)。结论血清蛋白质谱图存在差异的6个不同质荷比蛋白质组成的蛋白质组有可能成为前列腺癌筛查的血清标志物。质荷比为10268Da的蛋白指纹峰有助于区分前列腺癌患者与前列腺增生患者和正常人群。Objective To screening serum biomarkers in patients with prostate cancer with protein fingerprints technique. Methods The serum protein profiles were examined with surface enhanced laser desorption/ionization time-of-flight mass spec- trometry (SELDI-TOF-MS) technique in 22 patients with prostate cancer (PC) and 26 patients with benign prostatic hyperplasia (BPH) and 25 normal controls. Results The expression of three proteins (3 165Da, 3 982Da and 4 291Da) in PC group (9.794 ± 4.449, 3.968 ± 2.247 and 11,020 ± 6.069) was significantly higher than that in normal controls (5.062 ± 2.245, 1.823 ±1.432 and 6.525 ± 5.270); while the expression of two proteins (2 747Da and 10 268Da) in PC group (22.390 ± 12.179 and 0.953 ± 0.338) was significantly lower than that in normal controls (42.231 ± 16.970 and1.932 ± 0.864) and one protein (10 268Da) in PC group (0.953 ± 0.338) was lower than that in BPH (1.314 ± 0.493) (all P〈0.05). However, there was no difference in pro- tein profile between BPH and control groups (P :〉0.05). The expression of protein 3 960Da (3,447 ± 1.434) in PC with positive PSA (〉4μg/L) was higher than that in BPH with positive PSA (1.473 ± 0.971), but the expression of protein 10 268Da (0.847 ± 0.408) in PC with positive PSA was lower than that in BPH with positive PSA (1.538 ± 0.182) (both P〈0.05). Conclusion Six serum proteins in PC patients are abnormally expressed which may be used as serum markers of prostate cancer screening,
