摘 要:含有二硫键(S-S)的小分子二丙烯酰胺与氨基化合物发生Michael加成反应,可以聚合成含有多重二硫键的聚氨胺(SS-PAAs)。由于主链上含有对细胞氧化还原环境具有响应性的二硫键,SS-PAAs能够被细胞内的谷胱甘肽降解成为小分子链段,从而在保持基因转染效率的前提下大大降低对细胞的毒性。因此,SS-PAAs有望成为新一代高效安全的转基因载体。本文综述了含有多重二硫键的SS-PAAs这一类新型载体的合成、表征、应用等方面的研究进展,并展望了下一步的研究方向。Poly(amido amine)s with multiple disulfide linkages(denoted as SS-PAAs) could be synthesized from the monomers such as bisacrylamides and amines with disulfide bonds through Michael additional copolymerization.In cells,the SS-PAAs can be easily degraded into small molecular segments by glutathione,as the disulfide bonds are easily broken in reduction condition.This feature sharply minimizes the toxicity of SS-PAAs,but still maintains the high transfection efficacy.Therefore,the SS-PAAs have a great potential to be developed as a new type of efficient and safe gene vector.In this review,we summarized the progress of the research on SS-PAAs,including synthesis,characterization and the applications in biomedical fields during recent years.Further research directions are also proposed.
