摘  要:目的评价再灌注后多次给予富氢液对大鼠全脑缺血再灌注损伤的影响。方法成年雄性SD大鼠,随机分为3组:假手术组(S组)、脑缺血再灌注组(I/R组)和富氢液多次给药组(H组)。I/R组和H组采用四血管阻塞法制备全脑缺血再灌注损伤模型。H组大鼠于再灌注即刻及再灌注后2、4、6h分别腹腔注射0.6mmol/L富氢液5ml/kg。I/R组大鼠给予等容量的生理盐水。再灌注24h时,测定海马丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的含量;再灌注72h时,尼氏染色观察海马组织形态学变化。结果与I/R组比较,H组海马组织MDA、TNF-α、IL-6含量降低(P<0.05),尼氏染色见海马CA1区神经元、尼氏小体数目增多,细胞形态趋于正常。结论再灌注后多次给予富氢液可减轻大鼠全脑缺血再灌注损伤,其机制与抑制脂质过氧化反应和炎性反应有关。Objective To investigate the effect of hydrogen-rich saline given several times during reperfusion on the rats with transient global cerebral ischemia-reperfusion.Methods Adult male Sprague-Dawley rats were divided into 3 groups at random:sham operative group(S),cerebral ischemia-reperfusion group(I/R) and hydrogen-rich saline group(H).Rats in I/R and H all suffered global cerebral ischemia and reperfusion(I/R) with a four-vessel occlusion model.Rats in H group were injected intraperitoneally 0.6 mmol/L hydrogen saline respectively at immediate moment and 2h,4h,6h of reperfusion,5ml/kg;and rats in I/R group were injected the same volume normal saline.At 24h of reperfusion,rats' hippocampus were detected for malondialdehyde(MDA),tumor necrosis factor-α(TNF-α) and interlukin-6(IL-6) concentrations.At 72h of reperfusion,rats' hippocampus were stained with Nissl staining.Results Compared to I/R group,the concentrations of MDA,TNF-α and IL-6 were significantly reduced(P0.05),and Nissl staining showed that the number of neurons and Nissl bodies in hippocampal CA1 were increased and the cell morphology tended to become normal.Conclusion Hydrogen-rich saline given several times during reperfusion could reduce the injury of the brain against global cerebral ischemia-reperfusion in rats.The mechanism is related to the inhibition of oxidative stress and inflammatory responses in the hippocampus.
