摘 要:目的探讨紧密连接蛋白occludin及ZO-1在食管鳞状细胞癌及手术切缘正常食管黏膜组织中的表达及其临床意义。方法采用组织芯片和免疫组化法检测49例食管鳞状细胞癌患者的癌组织及手术切缘正常食管黏膜组织(距肿瘤组织>5 cm)中occludin和ZO-1的表达,分析食管鳞状细胞癌癌组织中occludin、ZO-1表达与肿瘤的增殖、侵袭和转移之间的关系。结果食管鳞状细胞癌组织中occludin及ZO-1的阳性表达明显低于手术切缘正常食管粘膜组织(P<0.01),且癌组织中occludin、ZO-1的表达与其分化程度及淋巴结转移有关(P<0.05)。结论食管鳞状细胞癌的occludin和ZO-1的表达下调或缺失与肿瘤细胞的增殖、侵袭和转移有关,在食管癌的进展中起重要作用。Objective To investigate the relationship between the tight junction proteins(occludin,ZO-1) expression and clinicopathologic parameters in human esophageal squamous cell carcinoma(ESCC).Methods The proteins expression of occludin and ZO-1 in human esophageal cancer tissues and their corresponding normal mucosa were detected by tissue microarray and immunohistochemical techniques.Results Expression level of occludin and ZO-1 decreased significantly in ESCC tissues compared with their adjacent normal esophageal epithelium(P 0.01).Furthermore,the expression of occludin and ZO-1 showed obvious correlation with degree of differentiation of esophageal carcinoma and lymph node metastasis(P〈0.05).Conclusion Decreased expression of occludin and ZO-1 may play an important role in the carcinogenesis of esophagus.
