摘  要:目的探讨二氧化碳(CO2)气腹对膀胱癌细胞黏附分子表达的影响。方法体外培养膀胱癌T24细胞,以15mmHgCO2压力,作用于癌细胞2、4和6h,以免疫细胞化学法检测各组T24细胞株E-钙调素,整合素α6β4、α2β1,ICAM-1及CD44V6等蛋白的表达。结果体外模拟CO2气腹对T24细胞体外黏附细胞分子表达有影响:ICAM-1、CD44V6表达升高(均为P<0.01);并与作用时间呈相关(均为P<0.05);E-钙调素的表达减少(P<0.01),随CO2气腹作用时间呈负相关(P<0.05)。整合素α6β4、α2β1的表达明显改变(均为P>0.05)。结论 CO2气腹环境可引起体外培养的膀胱癌细胞黏附细胞分子表达改变,这可能促进癌细胞扩散转移的能力。[Objective] To investigate the effects of carbon dioxide(CO2) pneumoperitoneum on cell adhesion molecules expression of human bladder carcinoma cells.[Methods] The cultured T24 cell line were incubated in vitro with CO2 insufflations at pressures 15 mmHg,and the time of treatment for each pressure were 2 h,4 h and 6 h respectively.The changes of adhesion molecules(E-cadherin,integrinα6β4 and α2β1,ICAM-1,CD44) in T24 cell line were detected by immunohistochemistry method respectively.[Results] The CO2 pneumoperitoneum were the factor influencing some adhesion molecules of the T24 cells.The expression of ICAM-1,CD44 were found increased(P 0.01,respectively),and there was a positive correlation with the CO2 treatment time(P 0.05,respectively).The expression of E-cad was decreased,and it was negative correlated with treatment time.The expression of integrin α6β4 and α2β1 were found no statistically significant change respectively(P 0.05).[Conclusion] Carbon dioxide pneumoperitoneum can make change on some adhesion molecules' expression,and that may increase the capability of invasion and metastasis in the laparoscopic therapy on bladder carcinoma.
