摘  要:目的:优化芡实多糖的水提醇沉工艺。方法:以苯酚-硫酸法测定多糖的含量,以多糖得率和糖含量为指标,选取提取温度(A)、提取时间(B)、固液比(C)、提取次数(D)4个因素设计正交试验,采用单因素实验考察醇沉浓度的影响。糖含量的绿多糖,以蛋白,并结果:在实验影响因素中,影响程度从大到小依次为A>D>B>C,最佳提取方案为:A1B2C3D3,即在70℃下,固液比1∶20时,提取2.5h,连续提取4次。最佳醇沉浓度为90%。结论:本实验优选出的工艺稳定可行,可以为进一步研究芡实提供参考。Objective:To optimize the extracting technique of polysaccharides from Semen Eurylie.Methods:The contents of polysaccharide were determined by Phenol-sulfuric acid colorimetry.With the yield and contents of polysaccharides as index,the effects of extraction temperature(A),extraction time(B),the ratio of solid to liquid(C),the times of extraction(D)were considered using orthogonal experimental design.The effect of ethanol concentration was considered using single factor test.Results:The factors of effecting on the extraction of polysaccharides were in the order of ADBC.The optimum extraction conditions were 20 times of water,refluxing and extracting for 4 times at 70℃,and 2.5h for each time as A1B2C3D3,the optimum ethanol concentration was 90%.Conclusion:The optimized extracting technique of polysaccharides is stable and reasonable,and can be used as reference of further studies on Semen Eurylie.
