摘 要:目的探讨脂肪源性干细胞(ADSC)移植对大鼠脑缺血再灌注损伤区炎症递质多聚腺苷酸二磷酸核糖聚合酶(PARP)和核因子-κB(NF-κB)表达的影响。方法采用线栓法复制大鼠大脑中动脉缺血再灌注模型,将动物随机分为假手术组、模型组和ADSC组。ADSC组经尾静脉注射PKH-26标记的ADSC细胞悬液(0.5ml,细胞密度为4×106个/ml),观察各组大鼠的神经功能损害评分,采用蛋白质印迹法和PCR分别检测半暗带区NF-κB和PARP蛋白及其mRNA的表达水平。结果 ADSC移植后可见PKH26标记的ADSC在半暗带区有表达。与模型组比较,ADSC组神经功能损害评分显著降低(P<0.05),半暗带区PARP蛋白、NF-κB蛋白和mRNA表达水平均显著下降(P<0.05)。结论脂肪源性干细胞移植可通过下调炎症递质NF-κB和PARP的表达抑制炎性反应,从而促进脑缺血损伤神经功能的恢复。Objective To investigate the effects of transplantation of adipose-derived stem cell (ADSC) on the expression of poly adenosine diphosphate ribose polymerase (PARP) and nuclear factor-κB(NF-κB) after cerebral ischemia-reperfusion in rats. Methods The rat model of middle cerebral artery occlusion was established. Rats were randomly divided into sham operation group, model group and ADSC group. 4×106 PKH-26 labeled ADSC were infused into the caudal vein in ADSC group. Neurological severity scores (NSS) was then determined. The expression of NF-κB and PARP in infarction region were determined by Western-bolt and quantitative real-time RT-PCR. Results PKH-26 labeled ADSC were found in ADSC group.Compared with the model group,the NSS in ADSC group was significantly decreased(P0.05),and the protein and mRNA expression level of PARP and NF-κB in penumbral area were significantly increased(P0.05). Conclusion ADSC transplantation can promote functional recovery of cerebral ischemia reperfusion rats. The effect may be related to the refraining inflammatory reaction by decreasing the expression of PARP and NF-κB.
