摘 要:目的探讨PRL-3的过表达或敲低对结直肠癌细胞相应的生物学特性的影响。方法常规进行细胞培养,应用倒置相差显微镜、扫描电镜观察细胞结构;应用运动小室实验检测PRL-3对结直肠癌迁移能力的影响;应用细胞粘附实验测定细胞在I型胶原上的粘附性。结果粘附实验结果表明,与SW480/EGFP/mock和SW480/EGFP细胞相比,SW480-EGFP-PRL-3细胞的粘附能力增强,而SW480-PRL-3-KD1细胞的粘附能力减弱,差异有统计学意义(F=22.013,P<0.01)。运动小室实验证明,SW480/EGFP、SW480-EGFP-PRL-3、SW480/EGFP/mock及SW480-PRL-3-KD1细胞的运动能力差异有统计学意义(F=21.368,P<0.01),说明PRL-3与结直肠癌细胞的运动迁移能力有关。结论 PRL-3基因是结直肠癌细胞粘附、迁移的促进基因。Objective SW480 cells with stable over-expression or knock-down of PRL-3 gene were established.Effects of PRL-3 on the biological behaviors of SW480 cells were studied. Methods Effects of PRL-3 overexpression and knock-down on cell motility and migration were assessed by in vitro transwell chamber and adhesion assay.Morphological changes of colorectal cancer cells after PRL-3 overexpression or knock-down were studied by scanning electron microscope and inverted phase contrast microscope. Result The result of in vitro migration assay showed that SW480-PRL-3-KD1 cells had significantly reduced invasiveness and motility as compared with SW480-EGFP-PRL-3 and SW480/EGFP cells. Conclusion PRL-3 gene plays an important role in adhesion and migration of CRC.
