槲皮素对前列腺癌细胞的放疗增敏性研究

摘　　要：目的观察槲皮素对人前列腺癌细胞株PC-3的体外放射增敏作用,并探讨其可能机制。方法应用细胞克隆形成法研究槲皮素对PC-3细胞的毒性和放射增敏作用,westen-blot法检测survivin蛋白的表达。结果槲皮素对PC-3细胞有一定的毒性,随着药物浓度增加而增加,用0.3mg/ml的槲皮素作用于PC-3细胞3、6、12、24小时后,各组均可见放射增敏效应,相应的放射增敏比SERDo分别为1.22、1.58、1.76、1.85。单独药物组和单独照射组对survivin蛋白的表达的影响不明显,药物加照射组明显可以抑制survivin蛋白的表达,与对照组比较,有统计学意义(P<0.05)。结论槲皮素对PC-3细胞的毒性呈剂量依赖性,放射增敏作用明显,随药物作用时间延长而增强。其机制可能与抑制survivin蛋白表达有关。Objective To investigate the radiosensitization of quercrtin on PC-3 cells and its mechanism.Methods The cytotoxicity and radiosensitization of quercrtin were investigated by using clongenic assay on PC-3 cells in vitro,and the expression of survivin protein were evaluated by western blot.Results The cytotoxicity of quercrtin was in a dose-dependent manner.With quercrtin exposed to PC-3 cells for 3、6、12 and 24 hours,the corresponding SERDo was 1.22、1.58、1.76 and 1.85.The combination of 0.3mg/ml quercrtin with 2Gy radiosensitization could significantly inhibit the expression of survivin protein,but 0.3mg/ml quercrtin or 2Gy radiosensitization could not.Conclusion The cytotoxicity of quercrtin was in a dose-dependent manner.Quercrtin has a radiosensitizing effect,and the radiosensitization is dependent on time.Survivin maybe involved in it.
