摘　　要：目的检测不同病程糖尿病大鼠视网膜组织中促红细胞生成素(erythropoietin,EPO)的表达部位并观察其动态改变,探讨EPO在糖尿病视网膜病变发生发展中的作用。方法选用健康SD大鼠50只,随机分成5组,每组10只,其中对照组30只(实验开始组10只,3个月对照组和6个月对照组各10只)腹腔注射枸橼酸缓冲液,实验组20只(3个月组和6个月组各10只)腹腔注射链脲佐菌素(streptozotocin,STZ),建立糖尿病动物模型,分别于3个月和6个月时摘除眼球,免疫组织化学技术检测EPO在视网膜组织中的表达部位及表达量的变化。结果实验组SD大鼠视网膜组织EPO的表达随着病程的延长而增强,表达部位主要在神经节细胞层、神经纤维层、内丛状层、外丛状层,内、外颗粒层亦有少量细胞表达。结论 EPO与糖尿病视网膜病变的发生、发展密切相关。Objective To investigate the expression of erythropoietin（EPO） in the retina of diabetic rats and its association with diabetic retinopathy（DR）.Methods Diabetes mellitus（DM） was induced by injection of streptozotocin（STZ） in 20 SD rats and other 30 healthy SD rats served as controls.Ten rats in DM or control groups were sacrificed and the eyeballs were enucleated at 180d and 360d of the experiment,respectively.The histological changes were examined with H-E staining,and EPO expression was evaluated by immunohistochemistry.Results The expression of EPO was enhanced with the progress of DR and was mainly found in ganglion cells layer,nerve fiber layer,inner and outer plexiform layer,seldom in internal and outer nuclear layer.Conclusion EPO might be related to the pathogenesis and progress of diabetic retinopathy.
