摘 要:目的探讨MDR-1基因骨髓造血干细胞移植对大鼠肝移植术后复发转移的影响。方法选用雄性Wistar大鼠30只为肝移植供体,另选雌性SD大鼠30只为异基因肝移植受体(将HCCLM6肿瘤组织原位接种于大鼠肝脏,10 d后行肝移植,2周后处死大鼠)随机分为两组,A组MDR-1基因骨髓造血干细胞移植联合肝移植组,B组为单纯肝移植组。比较两组大鼠肝移植术后2周生存率、肝内复发和肝外转移情况。结果术后两周,A组大鼠存活14只,B组存活10只(P<0.05)。两组大鼠肝内复发率均为100%,但复发瘤大小差异有统计学意义(P<0.05),A组为(323.23±29.56)mm3,B组为(692.73±142.56)mm3。A组肺转移率为22%,B组为70%,A组淋巴结转移率为15%,B组为50%,差异均有统计学意义(P<0.05)。结论 MDR-1基因骨髓造血干细胞移植对大鼠肝移植术后复发转移有一定抑制作用。Objective To investigate the effect of MDR-1 gene bone marrow hematopoietic stem cell transplantation on relapse and metastasis of liver cancer in rats after liver transplant.Methods Thirty male Wistar rats were used as liver donor and 30 female SD rats as recipients for allogeneic liver transplantation.HCCLM6 tumor tissue was inoculated in the liver of recipient rats and liver transplantation was performed after 10d.The recipient rats were randomly divided into group A(MDR-1 gene bone marrow hematopoietic stem cell transplantation combined with liver transplantation) and group B(liver transplantation).The rats were sacrificed 2 weeks after liver transplantation;the survival rate,cancer recurrence and metastasis rate after liver transplantation were observed and compared between two groups.Results Two weeks after liver transplantation,14 rats survived in group A and 10 in group B(P0.05),with the recurrence rate of 100% in both groups.The size of recurrent tumor in group A was significantly smaller than that of group H [(323.23±29.56)mm3 vs(692.73±142.56)mm3,P0.05].There were significant differences in pulmonary metastasis rate(22% in group A and 70% in group B) and lymph node metastasis rate(15% in group A and 50% in group B) between two groups(P0.05).Conclusion MDR-1 gene bone marrow hematopoietic stem cell transplantation can inhibit liver cancer recurrence and metastasis after liver transplantation in rats.
