摘 要:目的:研究温热作用在体外对人胆囊癌GBC-SD细胞增殖的影响。方法:体外培养的胆囊癌GBC-SD细胞,给予43℃热作用1h,以37℃正常培养温度为对照,分别收集作用0、6、12h后的细胞。应用MTT、免疫细胞化学和Western-blot等技术,检测GBC-SD细胞经温热作用后增殖活性和PCNA蛋白表达的改变。结果:43℃热作用1h对胆囊癌细胞具有较明显的抑制效应,可有效抑制PCNA的表达。结论:43℃温热作用1h是抑制胆囊癌GBC-SD细胞增殖活性的有效热剂量。Objective:To investigate the effect of hyperthermia on human gallbladder carcinoma cell lines in vitro.Methods: Human cultured gallbladder carcinoma cell GBC-SD was heated for 1 hour at 43℃.Then 0h,6h,12h later GBC-SD cells were collected respectively.The effects of hyperthermia on the proliferation of GBC-SD cells were evaluated by MTT assay.Expression of proliferating cell nuclear antigen(PCNA) was detected with immunocytochemical technique and Western-blot.Results: After lh heated at 43℃,the proliferation of gallbladder carcinoma GBC-SD cell was inhibited.Levels of PCNA were obviously down regulated in hyperthermia group compared with control group.Conclusion: One hour moderate hyperthermia treatment at 43℃ is the effective heat-dosage to inhibit human gallbladder carcinoma GBC-SD cell proliferate.
