摘  要:目的:探讨RUNX3、DNMT1在人胃癌组织中的表达,分析其与胃癌临床病理特征及Hp感染的关系。方法:取70例手术的胃癌及癌旁胃黏膜组织,逆转录聚合酶链反应(RT-PCR)法检测RUNX3及DNMT1基因的mRNA水平,免疫组化分别检测蛋白的表达水平。结果:胃癌组织中RUNX3的表达显著下调,DNMT1表达明显增高,RUNX3表达下调与肿瘤分化程度及淋巴结转移密切相关(P﹤0.05)。RUNX3下调的胃癌组织中DNMT1的表达明显高于RUNX3正常的组织(P<0.05),在Hp阳性的胃癌组织中RUNX3的表达明显低于Hp阴性的胃癌组织(P<0.05)。结论:RUNX3基因和DNMT1基因可能参与胃癌的发生发展,Hp感染在一定程度上参与RUNX3基因失活。Objective:To investigate the expression of RUNX3 and DNMT1 in human gastric cancer,and to find out the clinical significance and the effect of Helicobacter pylori(Hp) infection.Methods: Reverse transcriptase-polymerase chain reaction(RT-PCR) and immunohistochemical method were used to detect mRNA and protein expression level of RUNX3 and DNMT1 of primary gastric neoplasm and correspongding para-cancer tissues,which were obtained from surgically resection specimens of 70 patients.Results:The expression of RUNX3 in gastric cancer tissues was significantly decreased,but that of DNMT1 was obviously increased.The depression of RUNX3 was correlated with differentiation degree and lymph node metastasis(P〈0.05).In gastric cancer the expression of RUNX3 in the Hp-positive group was significantly lower than the Hp negative group(P〈0.05).Conclusion: RUNX3 gene and DNMT1 gene may be involved in the development of gastric cancer.To some extent,Hp infection may involved in inactivation of RUNX3 gene.
