摘  要:目的建立人晶状体细胞膜年龄相关性改变的体外实验模型。方法取3对完整晶状体(30、41、63岁),将每对中的1个晶状体经50℃加热20h,与对侧未加热的晶状体均行冷冻切片。将上述切片行6-酰基-2-二甲氨基萘染色,然后在双光子共聚焦显微镜下观察。测量距晶状体中心处不同位点的总极化图像灰度(即总极化值),以评估相应位点细胞膜流动性的变化。另取2个晶状体(28、47岁)的冷冻切片,将切片于50℃加热20h,观察加热与未加热切片的细胞膜流动性改变情况。结果完整晶状体经加热后,中央区域的细胞膜流动性较对侧未加热晶状体明显增加,加热后的晶状体切片较加热前细胞膜流动性亦明显增加。结论晶状体50℃加热是模拟晶状体细胞膜流动性年龄相关性改变的有效模型,可用于晶状体细胞膜流动性方面的研究。Objective To establish an in vitro experimental model of changes associated with human lens cell membrane aging. Methods Three pairs of intact lenses ( aged 30, 41,63 years) were acquired. One lens in each pair was heated at 50 ℃ for 20 hours. Frozen sections of all lenses were prepared, then stained with 6-dodecanoyl-2-dimethyl- aminonaphthalene (Laurdan) and observed under two-photon confocal microscope. Generalized polarization values at spe- cific distances from the center of lens were measured to assess cell membrane fluidity. Meanwhile, frozen sections from 2 lenses (aged 28 and 47 years) were heated at 50 ℃ for 20 hours. The membrane fluidity was compared between the heated and unheated sections. Results The membrane fluidity in the center of the lens was apparently increased in the heated lenses compared to the unheated ones, the same as the heated sections campared to the unheated sections. Conclusions Heating lens at 50 ℃ would be an effective model of lens cell membrane fluidity associated with age, which could be used for further researches.
