摘  要:目的为了探察证实黑质-纹状体DA能神经通路阻断对大鼠纹状体Mor和Calb阳性神经元及其蛋白质表达的影响。方法借助6-OHDA注射阻断前脑内侧束建立PD模型;应用APO诱导旋转、握力测试、Morris水迷宫方法检测模型大鼠的行为学改变;应用免疫组化染色和蛋白印迹技术检测PD模型大鼠纹状体Mor和Calb神经元形态结构和蛋白水平的变化。结果①阿扑吗啡诱导模型大鼠出现健侧方向的旋转。模型大鼠抓握悬吊时间明显较正常组长(P<0.05),其寻找和发现平台的时间也明显较正常组长(P<0.05)。②模型大鼠纹状体Mor阳性区域的面积明显小于正常组(P<0.05),Calb阳性区域面积的变化不明显(P>0.05)。③模型大鼠纹状体Calb蛋白的表达水平明显低于正常组(P<0.05)。结论黑质-纹状体DA能神经通路阻断可能通过突触传递机制影响其突触后神经元的形态结构、蛋白表达、生理机能。Objective To confirm the effects on Mor and Calb-positive neurons and the protein expression of Calb by blocking the nigrostriatal circuit.Methods The PD model was establisled by 6-OHDA injectin in nedial tereprain bundle;rotating induced by APO,grip strength test,the Morris water maze methods were used to detect the behavior change of rat model;immunohistochemistry and Western blot technique were used to detect the morpholoqical chauges of Mor an Calb-positive neurons and the expression of Calb.Results ① Apomorphine could induce the contralateral direction of rotation in rat model.Compared to norm group,Rat model took a remarkable increase in hang times compdned normal gruop(P0.05),the time of seeking and finding the platform,rat model was significantly longer than the control(P0.05).② Mor positive area of rat model was significantly smaller than that in the control group(P0.05),Calb positive area of rat model did not change significantly(P0.05).③ Calb protein expression level of rat model was significantly lower than the normal group(P0.05).Conclusion Blocking dopaminergic nigrostriatal circuit might affect morphdogical changes of postsynaptic neuron,protein expression,and physiological function through the synaptic transmission mechanism.
