摘  要:目的观察雷帕霉素对人胃癌SGC-7901细胞增殖的影响及其对缺氧诱导因子1α(HIF-1α)、血管内皮生长因子(VEGF)基因表达的影响。方法SGC-7901细胞经雷帕霉素处理后,以四甲基偶氮唑蓝(MTT)法检测细胞的增殖情况,逆转录聚合酶链反应(RT—PCR)检HIF-1α、VEGF基因的表达。结果体外雷帕霉素可抑制人胃癌SGC-7901细胞增殖,并下调人胃癌SGC-7901细胞HIF—1α、VEGF基因的表达。结论雷帕霉素可抑制人胃癌SGC-7901细胞增殖,并且可能是通过下调人胃癌SGC-7901细胞HIF-1α、VEGF基因的表达而起到抑制作用的。Objective To investigate the effect of rapamycin on the proliferation of gastric cancer cell line SGC - 7901, as well as hypoxia- inducible factor-1α( HIF-1α), vascular endothelial growth factor (VEGF)expression. Methods Gastric cancer cells were treated with rapamycin. Cell proliferation was detected by MTT assay. Semi - quanti- tative RT - PCR was applied to detect the expression of HIF - 1α and VEGF mRNA changes. Results Rapamycin inhibited proliferation of human gastric cancer SGC -7901 cells in vitro. Rapamyein in human gastric cancer cells reduced HIF- 1α, VEGF mRNA expression. Conclusion Rapamycin can inhibit proliferation of human gastric cancer SGC -7901 cells in vitro. Rapamycin may downregulate HIF- 1α and VEGF mRNA expression in human gastric cancer cells.
