摘 要:目的探讨茶多酚(TP)对人卵巢癌细胞株SKOV3增殖及NSA(DDP)敏感性的影响。方法用四甲基偶氮唑蓝(M1Tr)法检测单独使用不同浓度TP、DDP及低毒剂量TP联合DDP对人卵巢癌细胞株SKOV3的增殖抑制作用,分别计算抑制率、半数抑制浓度(IC50)及增敏倍数。结果TP、DDP单药均以剂量依赖的方式抑制卵巢癌细胞株SKOV3的增殖。低毒剂量的TP联合DDP作用于细胞后,增殖抑制率明显增加,IC50由单用DDP时的6.555mg/L下降到3.021mg/L,敏感性提高到单用DDP的2.17倍。结论TP对人卵巢癌细胞株SKOV3有生长抑制作用,低毒剂量TP与DDP联合应用可提高SKOV3细胞对DDP的敏感性,减少DDP的用量。Objective To study the effects of tea ployphenols (TP) on the proliferation of human ovarian cancer cell line SKOV3 and its sensitivity to eisplatin (DDP). Methods Ovarian cancer cell line SKOV3 was incubated sepa- rately with different concentrations of TP and DDP alone and a low - toxic dose of TP in combination with DDP. The in- hibitory rates of call proliferation were tested by MTT assay. The half maxial inhibitory concentration ( IC50 ) values and chemosensitivity sensitizing factors were calculated. Results When used alone, both TP and DDP inhibited the growth of SKOV3 cells in a dose - dependent manner. When combined with a low - toxic dose of TP, the inhibitory rates of call pro]iferation increased significantly. The IC50 of DDP on SKOV3 dropped from the original 6. 555 mg/L to 3. 021 mg/L, and the chemotherapeutic sensitivity of SKOV3 cells increased to 2.17 times of that of the DDP alone group. Conclusion TP has an inhibitory effect on the proliferation of human ovarian cancer cell line SKOV3, and can enhance the chemotherapeutic sensitivity of SKOV3 to DDP.
