摘 要:目的应用D-半乳糖(D—galactose,D—Gal)制备亚急性衰老动物模型,并对该模型进行评价。方法将16只sD大鼠随机分为2组,腹腔注射D-半乳糖建立亚急性衰老动物模型。采用Y型迷宫检测行为学变化,TUNEL染色观察海马CA1区神经元凋亡,免疫组化方法检测大鼠海马CA1区神经型-氧化氮合酶(neuronal nitric oxide synthase,nNOS)及半胱氨酸天冬氨酸蛋白酶9(cysteinyl aspartate—specific protease,caspase-9P35)的表达。结果与空白对照组相比,衰老模型组学习尝试次数明显增加[(77±5.2)次VS.(50±6.3)次],记忆正确次数明显减少[(3.74-0.8)次VS.(7.7±0.5)次],差异均有统计学意义(P〈0.01);衰老模型组较空白对照组海马CAl区TUNEL阳性细胞[(37.8±1.3)个/HPVS.(0.8±0.2)个/HP]及caspase-9P35表达[(37.6±1.8)个/HPVS.(6.2±1.3)个/HP]明显增加(P〈0.01),而nNOS表达[(6.8±1.5)个/HPVS.(22.8±3.6)个/HP]明显降低(P〈0.01)。结论应用D-半乳糖可成功制备亚急性衰老动物模型,体现了学习记忆障碍的行为学变化。Objective To evaluate the effect of an animal subacute aging model induced by D - galactose ( D - gal). Methods 16 rats were randomly divided into 2 groups. The subacute aging model was established by injecting in- traperitoneally with the D - gal. Y - electrical maze test was used to determine the ability of learning memory. TUNEL method was used to detect apoptosis of hippocampal neurons in the CA1 region. Immunohistochemistry were used to deter- mine the expression of nNOS and Caspase - 9P35 in hippocampus CA1 region. Results In the subacute aging model group, the ]earning times of "/- electrical maze were increased [ (77 -+ 5.2) vs. ( 50 + 6.3 ) ] and the memory times were decreased [ (3.7 +0.8) vs. (7.7 -+0.5) ~ than the control group(P 〈0.01 ). Compared with the control group, TUNEL - positive neurons [ ( 37.8 + 1.3 ) vs. (0.8 + O. 2) ] and the activity of Caspase - 9P35 [ ( 37.6 + 1.8 ) vs. (6.2 + 1.3 ) ] increased significantly in hippocampal CA1 subfield of the subacute aging model group ( P 〈 0.01 ). while the activity of nNOS [ ( 6.8 -+ l. 5 ) vs. (22.8 -+ 3.6 ) ] conspicuously were lower ( P 〈 0.01 ). Conclusion The subacute aging model can be established by intraperitoneal injection of by D - gal, reflect behavior variation caused by learning memory impediment, and imitate the character of senile dementia better.
